Background

The spinach stems of thorns (Amaranthus spinosus L.) are known as one of the plants that have a variety of benefits, especially in the health field. Rich in active compounds, such as saponins, flavonoids, tannins, and steroids/triterpenoids, which have the potential as antibacterial. This study focuses on testing the effectiveness of ethanol extract of thorn spilian rods on two harmful bacteria, namely Staphylococcus aureus and Escherichia coli, which often causes infection in humans. The increasing resistance of bacteria to antibiotics has made it essential to explore alternative treatments.

Research Purposes

The purpose of this study is to identify the characteristics and chemical content of simplicia and ethanol extract of thorn spinach stems. In addition, this study aims to evaluate the antibacterial activity of the ethanol extract of thorn spinach rods against Staphylococcus aureus and Escherichia coli. Understanding the antibacterial properties of natural compounds is crucial in developing new treatments for bacterial infections.

Research Methods

The thorn spinach is extracted using the method of maceration with 96% ethanol solvent. The first step is to do screening of simplicia and extracts to find out its chemical content. The antibacterial activity test was carried out by the diffusion method agar, using disc paper to determine the effective concentration and minimum inhibitory concentration (KHM) of the extract. The use of agar diffusion method is a widely accepted technique for testing antibacterial activity.

Research Result

The results showed that the spinach spinach spinach spinach has a water content of 5.96%, water soluble in 38.17%, the juice dissolves in ethanol 37.87%, total ash content 4.59%, and insoluble ash content that is not soluble acid of 2.07%. Both simplicia and thorn spinach extract contain flavonoids, alkaloids, saponins, and tannins, which are active compounds that provide antibacterial effects. The presence of these compounds suggests potential antibacterial activity.

Ethanol extract of thorn spinach stems shows significant antibacterial activity. At a concentration of 500 mg/ml, this extract is able to inhibit the growth of Staphylococcus aureus with a diameter of the inhibitory area of 14.12 mm, and against Escherichia coli with a diameter of the inhibitory area of 13.94 mm. KHM of this ethanol extract in both types of bacteria is detected at a concentration of 50 mg/ml, with a diameter of the inhibition area of 6.6 mm for Staphylococcus aureus and 6.3 mm for Escherichia coli. The results indicate that the ethanol extract has a strong antibacterial effect against both bacteria.

Conclusion

From the results of this study, it can be concluded that thorn spinach stems have the potential as an effective antibacterial agent against Staphylococcus aureus and Escherichia coli. This finding opens up the possibility to develop ethanol extract of thorn spinach rod as a natural ingredient in making antibacterial drugs, which can be used as an alternative in overcoming bacterial infections that are resistant to antibiotics. In addition, further research is needed to explore the clinical applications of this extract as well as its long-term safety and effectiveness. The discovery of new antibacterial agents is crucial in addressing the growing problem of antibiotic resistance.
